ENIGMA‑TB – “Understanding PE_PGRS, vital cell‑wall proteins promoting Mycobacterial survival in TB.”
The aim of this project is the structural and functional characterization of a selected panel of PE-PGRS proteins to better understand the mechanisms involved in the pathogenesis of Mycobacterium tuberculosis.
Hands-on skills
expertise in heterologous expression systems and downstream purification techniques
expertise in cloning methods and in analyzing DNA and proteins via electrophoresis (agarose gels, SDS‑PAGE)
protein crystallization for structural studies; circular dichroism (CD) spectroscopy to assess protein folding and secondary structure; light-scattering analyses (Dynamic Light Scattering, SEC‑LS) to determine protein size and aggregation; fluorescence-based assays and investigating protein–ligand interactions through isothermal titration calorimetry (ITC)
